SMDL Laborexpress 2000 offers an assay to detect and genotype the insertion/deletion (I/D) polymorphism in intron 16 of the human angiotensin-converting enzyme (ACE)-encoding gene by Real-Time PCR.
Angiotensin-converting enzyme (ACE) is a zinc metallopeptidase that is abundant on the surface of epithelial and endothelial cells. ACE plays a key role in two distinct physiological systems. It converts angiotensin I to angiotensin II, a strong vasoconstrictor, and breaks down bradykinin, a potent vasodilator. pressure by regulating fluid volume in the body.
Plasma levels of ACE vary widely among individuals, and several studies have shown that these variations may be due to an approximately 50% insertion/deletion (I/D) polymorphism in intron 16 of the ACE gene.
This polymorphism is determined by the presence (insertion, allele I) or absence (deletion, allele D) of a 289 bp Alu-repeat sequence. The following three different genotypes occur: I/I = homozygous for the insertion; I/D = heterozygous for the insertion and deletion; D/D = homozygous for the deletion.
Various studies have shown that the D allele is associated with higher plasma and tissue ACE levels, which are doubled in individuals carrying the D allele compared with individuals homozygous for the I (I/I) allele. D allele carriers also have an increased risk of endothelial damage and development of hypercoagulability and cardiovascular disorders. The association between the presence of the D allele and atherosclerosis, mainly due to altered carotid artery intima-media thickness (CIMT), has been confirmed by additional studies.
